Next, the specimen undergoes hematoxylin and eosin (HE) staining in jar number 2 for 1 minute and 20 seconds. This is followed by repeated washing steps in tank number 4 and differentiation in tank number 3 to enhance contrast. The process continues with unbluing in tank number 7 and additional washes to prepare the tissue for eosin staining in tank number 8. Subsequently, a series of alcohol gradient washes in tanks 9 through 13 progressively dehydrate the specimen, ending with clearing in xylene tanks 14 and 15. Each step is precisely timed, ranging from a few seconds to over a minute, ensuring thorough staining, differentiation, dehydration, and clearing for optimal microscopic evaluation.
